Sleep Traits Causally Affect the Brain Cortical Structure: A Mendelian Randomization Study

چکیده

Background: Brain imaging results in sleep deprived patients showed structural changes the cerebral cortex; however, reasons for this phenomenon need to be further explored. Methods: This MR study evaluated causal associations between morningness, ease of getting up, insomnia, long sleep, short and cortex structure. Results: At functional level, morningness increased surface area (SA) cuneus with global weighted (beta(b) (95% CI): 32.63 (10.35, 54.90), p = 0.004). Short SA lateral occipital (b 394.37(107.89, 680.85), 0.007. reduced cortical thickness (TH) paracentral (OR −0.11 (−0.19, −0.03), 0.006). TH parahippocampal −0.25 (−0.42, −0.07), No pleiotropy was detected. However, none Bonferroni-corrected values relationship structure five types traits met threshold. Conclusions: Our potentially show evidence a higher risk association neuropsychiatric disorders not only brain areas atrophy, but also an increase middle temporal zone. findings shed light on occurrence traits.

Mendelian Randomization Causal Analysis Adiposity as a cause of cardiovascular disease: a Mendelian randomization study

Background: Adiposity, as indicated by body mass index (BMI), has been associated with risk of cardiovascular diseases in epidemiological studies. We aimed to investigate if these associations are causal, using Mendelian randomization (MR) methods. Methods: The associations of BMI with cardiovascular outcomes [coronary heart disease (CHD), heart failure and ischaemic stroke], and associations o...

Causal Relationship between Adiponectin and Metabolic Traits: A Mendelian Randomization Study in a Multiethnic Population

BACKGROUND Adiponectin, a secretagogue exclusively produced by adipocytes, has been associated with metabolic features, but its role in the development of the metabolic syndrome remains unclear. OBJECTIVES We investigated the association between serum adiponectin level and metabolic traits, using both observational and genetic epidemiologic approaches in a multiethnic population assembled in ...

Inactive matrix Gla protein is causally related to adverse health outcomes: a Mendelian randomization study in a Flemish population.

Matrix Gla-protein is a vitamin K-dependent protein that strongly inhibits arterial calcification. Vitamin K deficiency leads to production of inactive nonphosphorylated and uncarboxylated matrix Gla protein (dp-ucMGP). The risk associated with dp-ucMGP in the population is unknown. In a Flemish population study, we measured circulating dp-ucMGP at baseline (1996-2011), genotyped MGP, recorded ...

5. Mendelian randomization

Associations between modifiable exposures and disease seen in observational epidemiology are sometimes confounded and thus misleading, despite our best efforts to improve the design and analysis of studies. Mendelian randomization, the random assortment of genes from parents to offspring that occurs during gamete formation and conception provides one method for assessing the causal nature of so...

Pleiotropy-robust Mendelian randomization.

Background The potential of Mendelian randomization studies is rapidly expanding due to: (i) the growing power of genome-wide association study (GWAS) meta-analyses to detect genetic variants associated with several exposures; and (ii) the increasing availability of these genetic variants in large-scale surveys. However, without a proper biological understanding of the pleiotropic working of ge...
